چکیده

Suspensions of dental plaque from eighteen children with high caries activity were hydrolyzed in vitro by a partially purified fungal dextranase isolated from Penicillium funiculosum NRRL 1768. The dextranase preparation appeared to lack 1,4glucanhydrolase and sucrase activity. The release of reducing substances from the plaque was measured by use of a 3,5-dinitrosalicylate reagent. Hydrolysis occurred at pH 5.1 but not at pH 7.0. Approximately 20 per cent of the total carbohydrate of plaque was consumed by the dextranase (2-3 per cent of the plaque dry weight). No isomaltose was found when the hydrolysates were analysed by a thin layer chromatography procedure which could detect as little as 0.1 pg of isomaltose. The findings suggest that the quantity of dextran in these plaque samples is low.

Detection and preliminary studies on dextranase-producing microorganisms from human dental plaque.

An enriched nutrient agar medium containing blue dextran has been utilized for the detection of dextranase-producing microorganisms in human dental plaque. When compared with the total viable anaerobic plaque flora, the proportion of these microbes in supragingival plaque from different individuals varied over a wide range. Preliminary characterization of some of the dextranase-producing microo...

Characterization of a dextranase produced by an oral strain of Actinomyces israelii.

A dextranase-producing, gram-positive, anaerobic, rod-shaped bacterium isolated from human dental plaque was identified as Actinomyces israeli. Although the extracellular dextranase (EC 3.2.1.11) formed by this microbe appeared to be constitutively produced, the bacterium did not utilize the reaction products as a carbon source during growth. A striking feature of the dextranase was the formati...

Dextran-induced aggregation in a mutant of Streptococcus sobrinus 6715-13.

A mutant of wild-type Streptococcus sobrinus 6715-13 has been isolated which resists aggregation by exogenous dextran. This variant is able to form adherent plaque deposits in vitro when cultured in the presence of sucrose and has dextranase activity. In these respects it is the complement of previously described isolates which are plaque formation defective but aggregation normal. Measurements...

In-Vitro Effect of Natural Honey on the Growth of Streptococcus sanguinis

Background and Aim:  Streptococcus sanguinis (S. sanguinis) is one of the effective bacteria in the formation of caries and periodontal diseases which are caused by dental plaque. Currently, the use of natural honey (NH) is considered as a method of reducing S. sanguinis and S. mutans in saliva, which results in reducing the dental plaque. The purpose of this study was to evaluate the effects o...

Optimization of Culture Conditions and Environmental Factors of Dextranase Enzyme Produced by Paecilomyces lilacinus

Dextranase (α-1,6-D-glucan, 6-glucanohydrolase; E.C.3.2.1.11) is an enzyme which hydrolizes the α1,6 glucosidic linkages in dextran, found in higher plants, mammalian tissues, fungi and bacteria [1]. The enzyme has several applications in medicine and sugar industry because of its hydrolizing capacity of dextran. The presence of dextran has many negative effects in sugar processing levels such ...
